大肠杆菌氨基糖苷类耐药菌株OMF膜蛋白分析(8100字)

摘  要:为了探明大肠杆菌对氨基糖苷类抗生素的耐药性与大肠杆菌外膜蛋白OMF的关系,本试验应用药物筛选试验六株对氨基糖苷类抗生素耐药的大肠杆菌菌株(分别为菌株A1、菌株B1、菌株A2、菌株B2、菌株A3和菌株B3)。然后应用十二烷基硫酸钠聚丙烯酰胺凝胶电泳(SDS-PAGE电泳)检测它们的OMF膜蛋白,从结构上分析对氨基糖苷类抗生素耐药的大肠杆菌膜蛋白与标准大肠杆菌膜蛋白的差异。结果显示出,在这六株对氨基糖苷类抗生素耐药的大肠杆菌中,与标准大肠杆菌外膜蛋白相比,其中菌株A1、菌株A2、菌株B2和菌株B3四株菌株的OMF膜蛋白都有缺失,其他两株菌株A3和菌株B1未见有蛋白带区的缺失。由此可证明,大肠杆菌对氨基糖苷类抗生素耐药的主要原因是由于OMF膜蛋白的缺失。

Analysis of Aminoglycoside resistant strains of E. coli OMF membrane protein

Abstract: For E. coli proved resistant to the aminoglycoside antibiotics and the relationship between the E. coli outer membrane protein of OMF. In this experiment, drug screening test six pairs of aminoglycoside antibiotic resistance of E. coli strains. Strains were A1, strain B1, strain A2, strain B2, strain A3 and strain B3. Application of sodium dodecyl sulfate and polyacrylamide gel electrophoresis (SDS-PAGE electrophoresis) testing their OMF membrane protein. From the structural analysis of aminoglycoside antibiotic resistance of E. coli membrane proteins and membrane proteins between the standard. The results show that, in the six pairs of aminoglycoside antibiotic resistance of E. coli, E. coli outer membrane protein, compared with the standard, which strains A1, strain A2, strains B2 and B3 strains OMF four strains of both membrane proteins are missing, the other two strains A3 and B1 strains there was no lack of protein bands.This can be seen that E. coli aminoglycoside antibiotics is mainly due to the lack of membrane proteins as OMF.
