多柱联用分离黄芪(当归)复方活性成分研究(12300字)
摘  要: 本文对黄芪/当归复方复杂体系的树脂吸附色谱分离方法进行了研究。将当归黄芪复方提取液以大孔树脂吸附,乙醇梯度洗脱的个部分直接以活性炭柱、聚酰胺柱、5号树脂柱等大孔树脂作为二级柱吸附后,以乙醇梯度洗脱。结果在聚酰胺柱,10%乙醇洗脱液中主要含1、2、4,30%乙醇洗脱液中主要含1、4,50%乙醇洗脱液和70%乙醇洗脱液中主要含4号物质;95%主要含5号物质;活性炭柱中,10%乙醇洗脱主要含1、2、5号物质;50%、70%、95%乙醇基本没有物质洗脱;5#树脂30%乙醇洗主要含1号物质;50%乙醇主要含3、4、5、6号物质,70%乙醇洗脱中主要含2、6号物质,95%乙醇主要含2、3号物质;实验结果表明,黄芪/当归复方中多组分经多柱联用分离后可有效实现纯化分离。

Study on separation of bioactive compounds group from a Prescription Containing Radix Astragali and Radix Angelicae Sinensis by functional adsorbent multi-column chromatography
Abstract: A chromatographic method coupled with different functions adsorbent for separating multiple components from Danggui-Bxue Tang was investigated elementarily in this work.. The extract of Danggui Buxie Decoction was absorbed with a macroporous resin and the adsorbates were eluted by a gradient. Elution of ethanol-water for primary separation of multiple compositions in Danggui Buxue Decoction. Then the very eluting fraction was adsorbed with different function resin including the polyamide, sephadex and maropcoporous resin and then the adsorbates were also eluted by a gradient solvent of ethanol-water for a fine isolation of multiple components in this formula. The results analyzed by HPLC indicated that the groups of compound 1,2 and 4, the groups of compound 1 and 4, the groups of compound 4 and the groups of compound 5 were eluted mainly by 10%, 30%, 50%, 70%, 95% ethanol, nespectively, from polyamide columm; The compound 1, 2 and 5 were eluted by 10% ethanol from active carbon column. The grounps of compound 1, the grounps of compound 3, 4 5and 6, the grounps of compound 2 and 6 and the groups of compound 2 and 3. were eluted mainly by 30%, 50%, 70%, 95% ethanol, respectively, from the column of microporons resin 5. So, the multiple components of Danggui Buxue Decoction could be easily separation.
