细胞因子SOCS3、Gfi-1、IL-2与凋亡因子Gadd45、Bcl2在小鼠衰老过程中的转录差异研究(13300字)
摘 要:本研究旨在研究小鼠的衰老过程中SOCS3、Gfi-1、IL-2与Gadd45、Bcl2基因在免疫系统中的表达。分别取三月龄与六月龄小鼠进行衰老造模处理与对照,同时取三组自然衰老年龄梯度的小鼠为材料。以从各组小鼠胸腺和脾脏中提取出来的总RNA为模板,通过反转录得到cDNA。以β﹣actin 作为内参基因,应用Gelpro32图像分析软件标定cDNA浓度,用PCR与电泳技术检测SOCS3、Gfi-1、IL-2与Gadd45、Bcl2基因在不同组间的转录差异。结果显示自然衰老年龄梯度组、造模对照组、衰老模型组中衰老相关基因SOCS3、Gfi-1、IL-2与Gadd45、Bcl2在免疫器官中出现了转录表达差异。小鼠的免疫系统衰老与SOCS3、IL-2和Gadd45这三个转录因子信号途径可能存在一定的相关性。

The Transcriptional Difference of Cytokine SOCS3、Gfi-1、IL-2 and apoptosis factor Gadd45、Bcl2 during Aging Process
Abstract: Expression of SOCS3, Gfi-1, IL-2 and Gadd45, Bcl2 gene in mouse immune organs during aging process was concerned in this study. Both three- and six-months aged mice were used as control and constructing aging model, additionally with three gradient aging mice groups. The total RNA was extracted as material, and then cDNA was synthesized through reverse transcription, Gene specific PCR reactions for SOCS3, Gfi-1, IL-2 and Gadd45, Bcl2 were performed using β-actin as internal standard genes, and the results were standardized by optical density using Gelpro32 image analysis software , The results showed that the genes have different expression levels in different mice groups, and the expression of SOCS3, IL-2 and Gadd45 might be involved in the aging process of mouse immune organs.
